[Small] Real-Time Observation of Protein Aggregation at Liquid-Liquid Interfaces in a Microfluidic Device

We introduce a droplet microfluidic device for real-time monitoring of protein aggregation at liquid-liquid interfaces, capturing adsorption, viscoelastic layer formation, and particle formation simultaneously. At high protein concentrations (>120 mg/mL), rapid aggregation and increased particle release occur under mechanical stress. These results underscore the need to address interface-induced instabilities in high-concentration antibody formulations and highlight microfluidic platforms as effective low-volume tools for the optimizationof therapeutic proteins and formulations.
