We analyzed the destabilizing interactions between antibodies and oleic acid, resulting from the degradation of polysorbate 80 in therapeutic formulations. Antibodies rapidly adsorb to the oleic acid–water interface, forming a viscoelastic layer that triggers particle formation upon rupture, with the propensity being protein-specific. Our findings further reveal that PS80 prevents adsorption only under certain conditions, and adding l-arginine effectively mitigates interfacial particle formation, highlighting the need for tailored strategies to mitigate aggregation in antibody formulations.
